Bioensayos en modelos celulares y animales de enfermedades humanas

Resumen: Laboratorio de cribado fenotípico o basado en dianas moleculares (receptores nucleares, cinasas, factores de transcripción). Análisis a tiempo real de la eficacia y toxicidad de pequeñas moléculas de nueva síntesis o medicamentos reposicionados en enfermedades raras.Modelos celulares 2D, 3D u organoides. Estudios de cinética y dosis-respuesta, a tiempo real sobre: proliferación celular, citotoxicidad, apoptosis, migración, invasión, metabolismo mitocondrial, angiogenesis, genes reporteros. fosforilación de proteínas. Sistema de análisis, de alto rendimiento Incucyte SX5 (hasta 6 placas de 96 pocillos, simultáneamente) de fenotipos celulares 2D, 3D u organoides. Modelos de enfermedades humanas en ratones modificados genéticamente: papel funcional de SOCS2 en enfermedades metabólicas (Diabetes tipo II, Diabetes Gestacional, trastornos de la gluconeogénesis y lipogénesis) o cáncer hepático. Summary: Phenotypic screening laboratory or based on molecular targets (nuclear receptors, kinases, transcription factors). Resumen: El procedimiento estrella para la regeneración de órganos y tejidos consiste en la sustitución de la zona dañada con trasplantes de células/tejidos autólogos y heterólogos. Sin embargo, éstos presentan importantes limitaciones como la reducida disponibilidad de los primeros y posibilidad de rechazo inmunológico de los últimos. Como alternativa, se ha propuesto el uso de materiales biocompatibles y biodegradables de origen natural y/o sintético que reproduzcan la organización microscópica natural de la matriz extracelular de los tejidos sanos y que provean de un microambiente óptimo para la adhesión, crecimiento, proliferación y diferenciación celular. Esta línea de investigación pretende utilizar los conocimientos básicos en el ámbito de la Biología Celular e Histología animal para diseñar el andamio que mejor simule la matriz extracelular del tejido/órgano diana (ej. matriz orientada para la regeneración de nervios periféricos; matriz no orientada para la regeneración de la piel) y testar su utilidad en la regeneración de tejidos con procedimientos in vitro e in vivo.

Drug Repurposing in orphan diseases.

Summary: Drug repurposing in rare diseases. Repositioning statins in the treatment of sarcomas and blood cancer. Drug repurposing aims to find existing EU or FDA-approved drugs for diseases. The basic idea of drug repurposing is to discover new useful activity for a distinct disease in an older clinically used drug or one that failed in later stages of development or disease develop resistance to drug. Farmacodinámica. Bioensayos para la identificación de actividades moduladores de receptores nucleares en el contexto de enfermedades metabólicas y cáncer

Resumen: Nuestro laboratorio se ha especializado en procedimientos para para la identificación de nuevos moduladores de las actividades de receptores nucleares (ER, GR, AR, LXR). Estos se llevan a cabo mediante aproximaciones químicas altamente eficiente basada en el uso de estructuras privilegiadas a partir de fuentes naturales o mediante estrategias sintéticas eficaces, estudios de modelización 3D-QSAR y Docking. Estas estrategias han sido extensamente validadas por su eficacia para descubrir agonistas, antagonistas o agonistas/antagonistas mixtos, y sus aplicaciones en enfermedades metabólicas o cáncer relacionadas con receptores nucleares. Fenotipado preclínico en modelos celulares y animales. Usando estas estrategias, nuestro laboratorio ha identificado numerosas moléculas de pequeño tamaño con actividades agonistas y/o antagonistas de los Receptores de Estrógenos con potencial interés en Diabetes, esteatosis hepática, enfermedades inmunoinflamatorias o cancer hormono dependiente. Aplicamos estrategias farmaco-toxicogenómicas en modelos representativos de enfermedades humanas para caracterizar el mecanismo de acción y predecir la toxicidad de los nuevos productos antes de definir su eficacia y/o toxicidad en in vivo.
Summary: Our laboratory has specialized in procedures for the identification of new modulators of nuclear receptor activities (ER, GR, AR, LXR). These are done through a highly efficient chemical approaches based on the use of privileged structures from natural sources or through effective synthetic strategies, 3D-QSAR modeling studies and Docking. These strategies have been extensively validated for their effectiveness in discovering agonists, antagonists, or mixed agonists/antagonists, and their applications in metabolic diseases or cancer related to nuclear receptors. Preclinical phenotyping in cell and animal models. Using these strategies, our laboratory has identified numerous small molecules with agonist and/or antagonist activities of Estrogen Receptors with potential interest in Diabetes, hepatic steatosis, immunoinflammatory diseases or hormone-dependent cancer. We apply pharmaco-toxicogenomic strategies in a disease-based model to characterize the mechanism of action and predict the toxicity of new products before defining their efficacy and/or toxicity in vivo.

Farmacodinámica. Bioensayos para la identificación de compuestos antitumorales-inhibidores de la via JAK-STAT-SOCS

Resumen: Las proteínas de la familia STAT (Signal Transducer and Activator of Transcription) son esenciales para regular la expresión de genes necesarios para el crecimiento y metabolismo de la células en todas las etapas del desarrollo de los mamíferos. En contraposición, la activación aberrante de las proteínas STAT resulta en el desarrollo de tumores, enfermedades inmunológicas y metabólicas. Esto justifica que estas proteínas sean importantes dianas farmacológicas y que se demande la identificación de moduladores (inhibidores o activadores) de sus actividades biológicas. El objetivo que perseguimos en el presente proyecto es el descubrimiento y desarrollo de nuevas moléculas anti-STAT (1, 3, 5) con potencial actividad antitumoral y/o inmunomoduladora. Para ello estamos realizando la evaluación farmacológica de una quimioteca (librería química) que ha sido diseñada para la identificación de inhibidores de las proteínas Jak/Stat. Esto se lleva a cabo mediante una aproximación altamente eficiente basada en el uso de estructuras privilegiadas a partir de fuentes naturales o mediante estrategias sintéticas eficaces, estudios de modelización 3D-QSAR y Docking. Esta estrategia ha sido extensamente validada por su eficacia para descubrir fármacos más selectivos, potentes y menos tóxicos. La capacidad de estos productos para modificar las actividades transcripcionales dependientes de STAT se evalúa, en primer lugar, utilizando líneas celulares transfectadas con sistemas reporteros de luciferasa. Usando estas estrategias, nuestro laboratorio ha identificado nuevos fármacos inhibidores del crecimiento de células de cáncer humano dependientes de la vía STAT. Paralelamente, en este proyecto se evalúan las moléculas más activas en relación con respuestas biológicas concretas como proliferación, apoptosis, migración, invasión celular, respuesta inflamatoria, o formación de colonias tumorales. Aplicamos estrategias farmaco-toxicogenómicas de alta resolución para caracterizar con precisión los mecanismos moleculares de los nuevos fármacos. Estos estudios se complementan con la evaluación toxicológica, antitumoral o antiinflamatoria en modelos de animales in vivo.
Summary: The STAT (Signal Transducer and Activator of Transcription) family proteins are essential for regulating the expression of genes necessary for cell growth and metabolism at all stages of development in mammals. In contrast, aberrant activation of STAT proteins results in the development of tumors, immunological and metabolic diseases. This justifies that these proteins are important pharmacological targets and that the identification of modulators (inhibitors or activators) of their biological activities is demanded. The objective that we pursue in this project is the discovery and development of new anti-STAT (1, 3, 5) molecules with potential antitumor and/or immunomodulatory activity. For this, we are carrying out the pharmacological evaluation of a chemical library (chemical library) that has been designed for the identification of inhibitors of the Jak/Stat proteins. This is done through a highly efficient approach based on the use of privileged structures from natural sources or through effective synthetic strategies, 3D-QSAR modeling studies and Docking. This strategy has been extensively validated for its effectiveness in discovering more selective, potent, and less toxic drugs. The ability of these products to modify STAT-dependent transcriptional activities is first assessed using cell lines transfected with luciferase reporter systems. Using these strategies, our laboratory has identified new drugs that inhibit the growth of human cancer cells dependent on the STAT pathway. In parallel, this project evaluates the most active molecules in relation to specific biological responses such as proliferation, apoptosis, migration, cell invasion, inflammatory response, or formation of tumor colonies. We apply high-resolution pharmaco-toxicogenomic strategies to accurately characterize the molecular mechanisms of new drugs. These studies are complemented by toxicological, antitumor or anti-inflammatory evaluation in in vivo animal models.

Farmacogenómica aplicada al descubrimiento y desarrollo de fármacos. DNA Microarrays (Plataforma Agilent) Bioinformática

Resumen: El laboratorio de Genómica del IUIBS dispone de una plataforma Agilent para el análisis de microarreglos de ADN que se ofrece a la comunidad científica de nuestro entorno académico y empresarial. La Fármaco-Toxicogenómica, definida como el estudio dinámico del transcriptoma y sus elementos regulatorios aplicado al descubrimiento del mecanismo de acción y toxicidad de los fármacos. En nuestro laboratorio disponemos de una Unidad de Genómica Funcional para abordar el estudio de la expresión de los genes, gran escala, en modelos temporales o dosis-dependientes. Esto ha permitido crear bases de datos extensas que están siendo usadas para descifrar las vías de señalización e identificar nuevos biomarcadores y dianas farmacológicas. Los biomarcadores que emergen de estos estudios pueden tener un valor pronóstico y pueden ser usados para predecir los resultados terapéuticos y la toxicidad de un fármaco. Esta aproximación metodológica, se está mostrando muy útil para acortar el tiempo de I+D+i+t de un fármaco, facilitar la identificación de combinaciones de medicamentos más potentes y menos tóxicas.
Summary: The IUIBS Genomics laboratory has an Agilent platform for the analysis of DNA microarrays that is offered to the scientific community of our academic and business environment. Pharmaco-Toxicogenomics, defined as the dynamic study of the transcriptome and its regulatory elements applied to the discovery of the mechanism of action and toxicity of drugs. In our laboratory we have a Functional Genomics Unit to address the study of gene expression, on a large scale, in temporal or dose-dependent models. This has allowed the creation of extensive databases that are being used to decipher signaling pathways and identify new biomarkers and drug targets. The biomarkers that emerge from these studies may have prognostic value and can be used to predict therapeutic outcomes and drug toxicity. This methodological approach is proving to be very useful for shortening the R+D+i+t time of a drug, facilitating the identification of more potent and less toxic drug combinations.
